摘要
目的建立识别蜂胶特征组分的二极管阵列检测器/高效液相色谱法。方法色谱柱为Zor-baxSB—C18(150mm×4.6mm,5.0μm),流动相为甲醇-0.1%(体积比)甲酸,梯度洗脱,柱温为30℃,流速为1.0mL/min,检测波长为280,320nm,并进行190~400nm全波长扫描。结果美国、陕西两种蜂胶图谱重叠率达到85%以上,40个特征组分相对峰面积存在差异。结论该法简便、准确、结果代表性强、重现性良好,为蜂胶质量控制标准的建立提供依据。
Aim To develop a method of multi-component quantitative analysis in propolis and the analytical mode based on reversed-phase HPLC with photodiode array detector. Methods The analysis were separated by using a Zorbax SB-C18 column (150 mm ×4. 6 mm, 5 μm) and methol-water(containing 0. 1% formic acid) as mobile phase with linear gradient elution. The flow rate was 1.0 mL/min and the column was set at 30℃. A photodiode array detector was monitored at the detection wavelengths of 280 nm,320 nm and UV spectra were recorded from 190nm to 400nm. Results The chromatographic overlap rate of two propolis samples (America, Shaanxi)was over 85% and the relative area of fourty characteristic components was of difference. Conclusion This method has advantages such as simplicity, selectivity and sensitivity as a routine procedure. It can be regarded as a method for the quality control of propolis.
