摘要
目的用反相高效液相色谱测定苹果渣中槲皮素的含量。方法采用Symmetry C18色谱柱(5μm,4.6mm×150mm),流动相为甲醇:0.4%磷酸(5:5),流速为1.0mL/min,柱温30%,检测波长为368nm。结果槲皮素在0.008~0.04ug呈良好线性关系(r=0.99904),回收率为98.93%,槲皮素的相对标准偏差为2.72%,最低检测限度为2pg。结论方法快速、简单,灵敏度高,样品测定结果为苹果渣综合利用提供理论依据。
Aim To adopt a RP-HPLC method for the determination of quercetin in apple pomace. Methods The content was determination on a Symmetry C is (5 μm ,4.6 mm × 150 mm)column, using methanol -0.4% phosphorin acid(50: 50) as the mobile phase ,and detected at UV 368nm. The column temperature was 30℃ and the flow rate was 1.0 mL/min. Results The method was proved to be linear in the range of 0. 008 - 0. 004 μg with a regression efficient of 0.999 04. The average recovery was 98.93% with RSD 2.72%. The minimum detection limit was 2pg. Conehmion The method was proved to be quick, simple and highly sensitive. The result of sample determination wtll supply reference for many-sided utilization of apple pomace.
