茶树新梢内源激素的HPLC分析及日变化 被引量:28

HPLC Analysis and Diurnal Variation of the Endogenous Hormones in the Shoots of tea Plants

摘要 研究了茶树新梢内源赤霉酸(GA3)、吲哚乙酸(IAA)、吲哚乙醛(IAAD)、脱落酸(ABA)和玉米素(Zeatin)等激素的HPLC分析方法。在ODS(C18)反相柱上,GA3、IAA、IAAD和ABA用甲醇—乙腈—KH2PO4缓冲液系统为流动相、Zeatin用甲醇—乙腈—磷酸缓冲液系统为流动相得到满意的分离,它们与相邻峰间的峰高分离度均在0.65以上。在紫外波长λGA3=210nm,λIAA、IAAD、ABA=280nm和λZeatin=254nm、灵敏度0.003aufs(GA3)和0.001aufs(IAA、IAAD、ABA和Zeatin)条件下,各种激素得到定量。GA3、IAA、IAAD、ABA和Zeatin的平均标样添加回收率分别为76.24%,89.69%,84.33%,90.96%和82.09%。茶树新梢内源激素含量在一天内有明显的变化,其高峰值均出现在13时左右。 A method was established for quantitative analysis of gibberellic acid(GA 3),indole 3 acetic acid(IAA),indole 3 acetaldehyde (IAAD),abscisic acid(ABA)adn Zeatin in tea plants by reverse phase HPLC.Seperation and purification of the endogenous hormones were obtained on an analytical ODS C 18 column(5 0×150mm),eluted with the solvent delivery systems methanol ethyl nitrile KH 2PO 4 buffer (GA 3,IAA,IAAD and ABA) and methanol ethyl nitrile phosphoric acid buffer (Zeatin) respectively.Their resolutions to the neiboring peaks were all above 0 65.They were detected by UV detector at 210 nm (GA 3,0 003 aufs),280 nm (IAA,IAAD and ABA,0 001 aufs) and 254 nm (Zeatin,0 001 aufs),and quantified by HPLC,with average recovery rates of 76 24%,89 69%,84 33%,90 96% and 82 09% respectively.Diurnal variations in levels of the endogenous hormones were also investigated by the HPLC method established.The contents fluctuated significantly daily and reached the maximum at about 1 pm.It is suggested that samples of tea shoots be collected from 10 am to 4 pm for the suitable quantification of the endogenous hormones.
