摘要
目的:建立高效液相色谱-蒸发光散射检测法测定桑白皮中1-脱氧野尻霉素(1-deoxynojifimycin,简称 DNJ)含量。方法:采用氨丙基键合硅胶为填充剂的色谱柱,流动相为乙腈-水(78:22),流速:0.8 mL·min^(-1),ELSD 检测器的漂移管温度为50℃,载气流速2.0×10~5 Pa。结果:DNJ 在2.12~21.20μg范围内呈良好的线性关系,r=0.9999;平均加样回收率为97.43%(n=6)。结论:该方法重现性好,灵敏度高,可有效控制桑自皮中 DNJ 的含量。
Objective:To develop an HPLC -ELSD method for determination of 1 -deoxynojiilmycin (DNJ) in Cortex Moil. Methods: This method used a Hypersil NH2 column and with a mobile phase consisted of acetonitrile - water(78: 22) ,0. 8 mL ·min^-1 as flow rate;Detector was ELSD , with an inlet temperature of 50 ℃, and its flow rate was 2. 0 × 10^5 Pa. Results:The linear range was 2. 12 -21.20 μg,r =0. 9999 ;The mean recovery was 97. 43% (n = 6). Conclusion:This method is of good reproducibility and sensitivity, and it can be used for the quality control of DNJ in Cortex Moil.
