HPLC determination of caffeine in compound quinine injection

目的:建立 HPLC 法测定复方奎宁注射液中咖啡因含量。方法:色谱柱为 PE Validated C_(18)(100 mm×4.6 mm,5μm),流动相为甲醇-1%醋酸溶液(用二乙胺调节 pH 至3.7)(35:65),流速为1.0 mL·min^(-1),检测波长为272 nm,采用外标法计算含量。结果:本法简便可靠、灵敏、准确,咖啡因在5.56～55.6μg·mL^(-1)范围内呈良好的线性关系(r=0.9998),精密度(RSD=0.26%,n=5)和重复性(RSD=0.27%,n=5)良好。结论:建立的定量方法专属性强,可用于复方奎宁注射液中咖啡因的含量测定。

Objective：To develop an HPLC method for determination of caffeine in compound quinine injection. Methods ：APE Validated C18（ 100 mm ×4.6 mm,5μm） column was used with a mobile phase consisted of mixture of methanol - 1% acetic acid（pH 3.7 ,adjusted by diethylamine） （35： 65）by flow rate of 1.0 mL ·min^-1 in this method. And UV detector wavelength was 272 nm. Calculation was processed by external standard method. Results： This method is simple, reliable, sensitive and accurate,in which caffeine has a good linearity （r = 0. 9998 ）with the concentration range from 5.56 -55.6 μg · mL^-1. However,this method also have better precision（RSD =0. 26% , n = 5 ） and good reproducibility （ RSD = 0. 27%, n = 5 ）. Conclusion： An accurate, simple, rapid qualitative method with good reproducibility to assay caffeine in compound quinine injection.
