摘要
目的测定盐酸万古霉素,盐酸去甲万古霉素血药浓度。方法采用离子对高效液相色谱法。色谱柱为Shimpack C18柱(150×6mm,5μm),加AlltechC18保护柱;流动相为乙腈-离子对缓冲液(115:885,pH3.1);流速1mL·min^-1;检测波长236nm,柱温40℃。结果盐酸去甲万古霉素在0.80~80.19μg·mL^-1;盐酸万古霉素在1.08~108.00μg·mL^-1浓度范围内呈良好的线性关系,两者低、中、高回收率在97%~110%之间,RSD在2%~6%之间。结论本方法简便可行,快速可靠。适合于临床血药浓度监测。
OBJECTIVE To determine the blood concentration of vancomycin and norvancomycin. METHODS HPLC was used. CH3CN-ion paired buffer( 115:885, pH3.1 ) was used as mobile phase on Shimpack C18 (150 × 6mm,5 μm)with column temperature at 40℃. The flow rate was 1mL· min^-1. Detection wavelength was 236nm. RESULTS The calibration curve was linear within the range of 0.80 - 80. 19μg · mL^-1 for norvancomycin and 1.08 - 108.00μg · mL^-1 for vancomycin. The average recovery within high and low blood concentration of norvancomycin and vancmyein were 97% - 110% with RSD among 2% - 6%. CONCLUSION This method was simple,rapid and suitable for the clinical monitor of norvancomycin and vancomycin.
