不同产地柴胡药材GC-MS指纹图谱研究 被引量:26

摘要 目的对不同产地柴胡药材的挥发油成分进行GC—MS指纹图谱比较研究。方法利用GC色谱指纹图谱的方法,以道地产区河北省主产的7批北柴胡药材为基准,对比不同产地柴胡药材的色谱指纹图谱。色谱条件:HP-1非极性柱,进样口温度250℃,检测器(FID)温度270℃,程序升温50-270℃,体积流量为0.9mL/min,分流比为10:1。质谱条件:电离方式为EI,离子源温度300℃,电子能量70eV。结果从7批道地产区柴胡药材挥发油GC指纹图谱中提取19个共有峰,用质谱分析鉴定出其成分,并对不同产地药材进行相似度的比较。结论柴胡挥发油气相色谱指纹图谱中各成分均得到了较好的分离,可作为柴胡药材质量评价的专属性的指纹图谱,并以此评价不同产地柴胡的品质。 Objective To apply GC-MS fingerprint to the comparison of volatile oils in Radix Bupleuri collected from different habitats. Methods The samples from various habitats were compared by application of GC fingerprinting. GC Methods were optimized in order to get high quality GC profile. The GC system consisted of Agilent 6890 and FID. The analysis was performed on an HP-1 column. The temperatures of sample vent and FID were 250 ℃ and 270 ℃, respectively. The column temperature was sequentially elevated from 50 ℃ to 270 ℃ at 3.5 ℃/min. The carrier gas was N2, and its flow rate was 0. 9 mL/min. The refractive ratio of distillation was 10 : 1. MS condition: ionization mode was EI; source temperature was 300℃, and electron energy was 70 eV. Results The GC fingerprints were established and the 19 common peaks were found and identified with GC-MS in volatile oil of seven batches of Radix Bupleuri. The comparison on similarity degree of the crude drugs from various habitats has been carried on. Conclusion A good fingerprint of Radix Bupleuri has been described. The method is simple and accurate with good reproducibility. The GC fingerprinting with specificity can be applied to estimate and control the quality of Radix Bupleuri.
