促智方对老年大鼠学习记忆减退改善作用的研究

摘要 目的:研究促智方对老年学习记忆减退大鼠海马突触素的影响。方法:用Morris水迷宫法筛选出老年学习记忆减退大鼠与老年学习记忆正常大鼠(SMN),将筛选出的大鼠分为促智方组(CUZHI-FANG)与老年学习记忆减退对照组(SMDc)。用免疫组织化学与图像分析技术,观察各组大鼠海马CA3突触素免疫活性。结果:促智方组突触素免疫活性明显高于SMDc组相应各区(P<0.01)。结论:促智方可能通过调节海马CA3神经元突触密度,改善学习记忆功能。 Objectives: To observe the effect of the herbal medicine CUZHIFANG on synaptophysin immunoreactivity in hippocampus of aged learning and momory impaired rats. Methods: Sixty male healthy rats (10 young and 50 old)were examined with Morris water maze. The old rats were divided into senile memory deficits rats (SMD) and senile memory normal rats (SMN). The SMD were further divided into CUZHIFANG group and SMD control group (SMDc). The synaptophysin (SYN) immunolabeling density was estimated in freezed sections of hippocampal formation of rats by the aid of image analysis system. Results: A significant increase (P 〈0.01) in the value of the calibrated optic density (COD) of synaptophysin immunoreactivity particles was found in the hippocampus CA3 in the CUZHIFANG group, compared with the SMDc group. Conclusion: CUZHIFANG improves learning and memory of aged rats in morphology. The probable mechanism of CUZHIFANG is by increasing hippocampal synaptic density in CA3.
