乳腺癌组织和癌旁组织MGMT和GST-π蛋白表达的流式细胞术分析 被引量:2

摘要 目的研究乳腺癌组织和癌旁组织O6甲基鸟嘌呤DNA甲基转移酶(MGMT)和谷胱苷肽S转移酶π(GSTπ)蛋白表达的临床意义。方法应用流式细胞术(FCM)分析MGMT和GSTπ蛋白表达。结果乳腺癌组织MGMT和GSTπ表达显著高于癌旁组织(P<0.05)。2种耐药相关蛋白在不同年龄、不同肿瘤大小、不同病理类型、不同临床分期和雌、孕激素受体阴性与阳性患者间的表达均无显著差异(P>0.05)。有淋巴结转移组患者MGMT与无淋巴结转移组患者比较无显著差异(P>0.05)。但GSTπ表达在有淋巴结转移组患者显著高于无淋巴结转移组患者(P<0.05)。相关分析显示,MGMT与GSTπ表达间无显著相关性(P>0.05)。结论初步研究结果提示,MGMT蛋白表达可能与乳腺癌预后无关,不能作为乳腺癌的一个预后指标。GSTπ表达与淋巴结转移状况有关,有可能成为预测乳腺癌预后的指标之一。 Objective Our purpose was to explore the clinical significance of MGMT and GST-π protein expression in breast carcinoma tissues and corresponding para-carcinoma tissues. Methods Flow cytometry was used to analyze the expression of MGMT and GST-π protein. Results The expression of MGMT and GST-π were higher in breast carcinoma tissues than that in corresponding para-carcinoma tissues(P〈 0.05). There were not notable differences of two drug resistance related protein expressions in different ages, different tumor sizes, different pathological types, different clinical stages, estrogen receptor or progestin receptor negative and positive patients (P〉0. 05). There was no notable difference of MGMT expression in lymph node metastasis negative and positive patients(P〉0. 05). The expression of GST-π was higher in lymph node metastasis positive patients than that in negative patients(P〈0. 05). The correlation analysis showed that there were no correlations between the expressions of MGMT and GST-π(P〉 0. 05). Conclusion The preliminary research results suggest that MGMT protein expression had probably no correlation with prognosis of breast carcinoma patients,it could not become a prognosis target of breast carcinoma. The expression of GST-π had related with lymph node metastasis status,it can become a target in forecasting breast carcinoma prognosis.
