摘要
目的:建立以高效液相色谱法测定家兔血浆中己酮可可碱缓释胶囊浓度的方法。方法:色谱柱为Hypersil C18,流动相为乙腈-磷酸盐缓7中液(25:75),流速为1ml/min,检测波长为274nm,柱温为30℃,内标为非那西丁,血样用二氯甲烷萃取。结果:血浆中己酮可可碱检测浓度在10~400ng/ml范围内与峰面积积分值线性关系良好(r=0.9995),平均回收率为90.01%(RSD=2.85%)。结论:本方法可用于己酮可可碱缓释胶囊血药浓度的测定。
OBJECTIVE: To establish HPLC method for the determination of plasma level of oxpentifylline delayed release capsule in rabbits. METHODS: The determination was performed Hypersil C18 column at room temperature (30℃). The mobile phase consisted of acetonitrile- buffer phosphate (25 : 75) with a flow rate of 1ml/min and detecting wavelength at 274nm. With phenacetin as internal standard, the blood sample was extracted with dichlormethane. RESULTS: The linear concentration range of oxpentifylline in plasma was 10~400ng/ml (r = 0.9995), and the average recovery was 90.01% (RSD = 2.85% ). CONCLUSION: The established method is suitable for the determination of oxpentifylline modified release capsule.
