摘要
目的测定不同周龄和超排前后雌性Fmr1基因敲除FVB小鼠和背景FVB小鼠血清生殖激素雌二醇(E2)、孕酮(P)、促黄体生成激素(LH)和促卵泡生成激素(FSH)含量,比较不同周龄和不同状态下敲除基因动物的血清性激素水平,研究敲除基因对动物的发育和生殖生理等方面的影响。方法用放射免疫分析法测定上述动物不同周龄和超排前后血清E2、P、LH和FSH的含量,测定Fmr1基因敲除小鼠不同周龄时的超排卵数,进行统计学分析。结果6周龄的雌性Fmr1基因敲除小鼠与FVB小鼠比较,LH、FSH值差异无显著性(P>0.05),E2的P值差异有显著性(P<0.05)。10周龄的两组动物间血清生殖激素水平和超排卵数均差异无显著性(P>0.05)。10周龄Fmr1基因敲除小鼠的超排卵数与6周龄动物比较差异有显著性(P<0.05)。结论敲除Fmr1基因对动物雌性激素的分泌有影响,该基因与动物的生殖生理有一定的关联。
Objective To assess and analyze the levels of estradial ( E2 ), progesterone (P), follicle-stimulating hormone(FSH) and luteinizing hormone (LH) in blood serum of Fmrl knockout mice and FVB mice before and after superovulation. Method The levels of E2, P, FSH and LH were measured by BIA and statistically analyzed. Results The levels of FSH and LH were not different between Fmrl knockout mice and FVB mice. The levels of E2 and P were higher in FVB mice ( P 〈 0.01 ). The levels of E2, P, FSH, LH and the amount of ova had no signitlcunt difference between Fmrl knockout mice and FVB mice at 10 weeks of age after superovulation ( P 〉 0.05), but the amount of ova of superovulated Fmrl knockout mice 10 weeks old was higher than those in 6 weeks old ( P 〉 0.05). Conchmion Fmrl gene may perhaps influence the E2, P, FSH and LH levels in serum . It should be taken into consideration in mouse reproductive physiological studies.
