血小板反应素Ⅰ在糖尿病大鼠心肌组织中的表达及意义

摘要 目的:研究血小板反应素Ⅰ(TSP-Ⅰ)在链脲佐菌素诱导的糖尿病大鼠模型心肌组织中的表达,探讨TSP-Ⅰ在糖尿病性心肌病发病机制中的作用。方法:成年Sprague-Dawley雄性大鼠20只,随机分为糖尿病组(DM组)和正常对照组(NC组),DM组用链脲佐菌素诱导产生糖尿病模型,NC组大鼠尾静脉注射pH 4.6、0.1mmol/L枸橼酸钠缓冲液。饲养4周后,麻醉处死,取出心脏,制做连续冰冻切片,对2组大鼠心肌组织进行免疫组织化学染色,观察2组TSP-Ⅰ在左心室心肌细胞中的表达差异。结果:DM组大鼠心肌间质纤维增生,心肌间微血管管壁增厚,TSP-Ⅰ表达阳性心肌细胞的数目及其光密度均较NC组明显增加(P<0.01)。结论:TSP-Ⅰ可能参与了心肌细胞肥大和纤维化以及心肌微血管病变等糖尿病心肌病的特征性病理改变。 Objective. To investigate the expression of thrombospondin- Ⅰ (TSP- Ⅰ ) in diabetic model induced by Streptozotocin, and its roll in the pathogenesis in myocardosis. Methods. Twenty adult male Sprague-Dawley rats were divided into two groups at random. In diabetic group, the animals were induced to diabetes by Streptozotocin injection, which was proved by blood and Urinary sugar level while the control was injected by 0.9% saline. After continue fed for 4 weeks, these animals were killed and the hearts were taken out. The freeze sections of these tissues were stained with TSP-Ⅰ immunohistochemistry method. Results. Compared with control group, We found the densities and numbers of TSP- Ⅰimmunostaining positive myocardial cell in left ventricle showed a significant increase in the heart of diabetic rats. Conclusion. The upregulation of TSP- Ⅰin the left ventricle myocardium in diabetic model may relate to pathologic changes of diabetic heart disease, such as hypertrophy, fibrosis of myocardial cell and microvessel dissepiment thickening.
