摘要
探讨PBX1基因转染人单核细胞系THP1后,对IFI16基因表达的影响。以携带人PBX1基因表达序列的复制缺陷型5型腺病毒载体(pDC315-PBX1-EGFP)转染THP1细胞后,采用实时定量PCR(RT-PCR)和Western blot技术分别在mRNA水平和蛋白水平检测PBX1和IFI16表达的变化。结果,pDC315-PBX1-EGFP转染THP1细胞后,PBX1基因的表达水平都增高,同时发现IFI16基因的表达水平降低。PBX1作为一种转录因子,能调节干扰素诱导蛋白IFI16的表达。
To investigate the influnce of the pre-B cell leukemia transcription factor 1 (PBX1) on the expression of the SLE-susceptible gene encoding for the inteferon inducible nueleoprotein 16(IFI16), the replication defective adeno virus type 5 vector carrying the expression sequences of human PXB1 gene (pDC315-PXB1-EGFP) was transfected to human monocytes THPI cells, and the effect of over-expresslon of PXBI on IFI16 was then evaluated by RT-PCR and Western blotting analysis. It was demonstrated that after the plasmld pDC315-PXB1 EGFP was transfeted to THPI cells, the expresxion of PBX1 mRNA and protein was significantly increased, and the that of IFI16 mRNA and protein decreased. It is concluded that over expression of PBX1 can down regulate the expression of IFI16.
