低氧诱导肾细胞癌786-O细胞HIF-1α的表达变化及意义

摘要 目的探讨缺氧条件下肾透明细胞癌786-O细胞缺氧诱导因子1α(HIF-1α)的表达变化及意义。方法786-O细胞在缺氧条件下培养不同时间(0、8、16、24h)后,RT-PCR法检测HIF-1αmRNA的转录情况;Westernblot法检测HIF-1α蛋白表达变化。结果常氧状态下HIF-1αmRNA和蛋白均有一定表达。缺氧处理一定时间(8、16、24h)后786-O细胞HIF-1αmRNA表达没有明显改变(P>0·05),而HIF-1α蛋白表达水平显著升高(P<0·01)。结论缺氧条件下786-O细胞HIF-1α表达变化主要在翻译水平而非转录水平。在蛋白水平对HIF-1α表达进行调控可作为肾透明细胞癌治疗的新靶点。 Objective To investigate the changes in the expression of hypoxia inducible factor 1α (HIF-1α) in renal clear carcinoma 786-O cells under different hypoxia phases and its significance. Methods 786-O cells were cultured under low oxygen tension for different durations (0, 8, 16 and 24 hour). The mRNA and protein expressions of HIF-1α in 786-O cells were detected by reverse transcription-polymerase chain reaction analyses (RT-PCR) and Western blot analyses respectively. Results mRNA and protein expression of HIF-1α could be observed in renal carcinoma 786-O cells under normal culture condition. After the hypoxia treatment (8, 16 and 24 hours), the mRNA level of HIF-1α did not changed significantly (P〉0. 05). However, the protein expression of HIF-1α was increased remarkably with correspondence to the different hypoxia time (P〈0. 01). Condusion The expression of HIF-1α in renal clear carcinoma was elevated under hypoxia. The regulation of HIF-1α in renal clear carcinoma may be on the translation level instead of transcription level under the different hypoxia conditions. It suggested that controlling the protein expression of HIF-1α may be a method to suppress the growth of renal clear carcinoma.
