摘要
目的:分析脂联素基因外显子2Gly15Gly(T45G)多态性与代谢综合征的相关性。方法:选择2003-10/2005-10在首都医科大学宣武医院高血压门诊就诊的高血压患者中具备代谢综合征所有组分者156例,另外选择同期行健康体检的健康者133例作为正常对照组,均签订知情同意书。采用聚合酶链反应-限制性片段长度多态性方法检测两组调查对象脂联素基因Gly15Gly的基因型,比较两组基因型和等位基因频率,并按年龄、性别进一步分组,分析两组不同年龄、不同性别间调查对象基因型及等位基因频率的差异。结果:纳入代谢综合征组调查对象156例,正常对照组133例,均成功进行了目的基因的扩增,无脱落。①两组调查对象的基因型及等位基因频率分布比较:正常对照组的TT,TG,GG基因型频率分别为51.9%,39.8%和8.3%;代谢综合征组分别为46.8%,42.3%和10.9%,两组比较差异均无显著性意义(P>0.05)。正常对照组的T,G等位基因频率分别为61.8%,28.2%;代谢综合征组分别为67.9%,32.1%,两组比较差异均无显著性意义(P>0.05)。②不同性别、不同年龄间两组调查对象的基因型及等位基因频率比较:进一步按照年龄、性别分组后,差异也未见显著性意义(P>0.05)。结论:脂联素基因外显子2Gly15Gly多态性可能与代谢综合征无明显相关性。
AIM: To analyze the association between adiponectin gene exon 2 Gly15Gly (T45G)polymorphisms and metabolic syndrome. METHODS: A total of 156 subjects in hypertensive clinic of Xuanwu Hospital, Capital University of Medical Sciences who had all the metabolic syndrome compositions and 133 corresponding healthy controls who had body examination at the same time were involved. They all singed the informed consent. The polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method was used to determine the genotype of adiponectin gene Gly15Gly in the two groups. Frequencies of genotype and allele were compared. The difference in the frequencies of genotype and allele was analyzed in different age and different sex. RESULTS: A total 156 in the metabolic syndrome group and 133 in the normal control group were undertaken to the amplification of objective gene, no drop-out. ①Comparison of genotype and allele frequency in the two groups: The frequencies of TT, TG and GG genotypes were 51.9%, 39.8% and 8.3% in the normal control group, 46.8%, 42.3% and 10.9% in the metabolic syndrome group, respectively. There was no significant difference between the two groups (P 〉 0.05). The frequencies of T and G allele were 61.8% and 28.2% in the normal control group, 67.9% and 32.1% metabolic syndrome group. There was no significant difference between the two groups (P 〉 0.05 ). ②Comparison of genotype and allele in different sexes and different ages in the two groups: There was no significant difference after grouping based on age and sex (P 〉 0.05). CONCLUSION: There may be no significant correlation between adiponectin exon 2 Gly15Gly polymorphism and metabolic syndrome.
