缺氧诱导因子-1α和血管内皮生长因子在乳腺癌中的表达及相关性 被引量:4

The expression of HIF-1α and VEGF and their relationship in breast cancer

摘要 目的本研究旨在探讨乳腺癌组织中缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)的表达;它们的内在相关性及其与乳腺癌生物学行为之间的关系。方法采用免疫组织化学S-P方法检测119例乳腺癌和10例乳腺良性肿瘤组织中HIF-1α和VEGF的表达。结果119例乳腺癌组织中HIF-1α和VEGF阳性表达率均为50.42%(60/119),显著高于乳腺良性肿瘤组织0/10;HIF-1α和VEGF的阳性表达率与乳腺癌患者临床分期、淋巴结转移情况和5年生存率密切相关(P<0.01);HIF-1α表达与VEGF呈正相关(P<0.01)。结论在缺氧状态下,乳腺癌组织中HIF-1α基因被激活,过渡表达HIF-1α蛋白,并通过VEGF的上调而刺激肿瘤新生血管的生成,使肿瘤耐受缺氧,促进乳腺癌的转移。HIF-1α和VEGF的表达与乳腺癌的生物学行为有关,可能成为预测乳腺癌侵袭、转移和预后的重要指标。 Objective This study was designed to examine the expression of HIF-1α gene and vascular endothelial growth factor (VEGF) protein in breast cancer, to find out whether there is significantly association among HIF-1α and VEGF and to reveal the relationship between them and the biological behavior of breast cancer. Methods Immunohistochemical streptavidin/peroxidase (SP) was used to examine the expression of HIF-1α and VEGF in 119 cases of breast cancer and 10 cases of benign breast tumors. Results HIF-1α and VEGF expression in breast cancer were beth 50.42% (60/119) significantly higher than that in benign breast tumors 0% (0/10). Positive expressiorrs of HIF-1α and VEGF were closely correlated with clinical stage, lymph node metastasis and 5 years survival rate of patients. The expression of HIF -1α was positively correlated with the VEGF protein. Conclusion The HIF-1α gene may be activated by hypoxia, and the overexpression of HIF-1α protein may induce tumor angiogenesis by activating the transcription of VEGF gene, suffeting hypoxia, promoting metastasis of breast cancer. The level of HIF-1α and VEGF related with biological behavior of breast cancer. They can be taken as useful marker for predicting invasion ,metastasis and prognosis in breast cancer.
