不同放射性浓度^(89)Sr诱导下乳腺癌MCF-7细胞的死亡模式

摘要 目的探讨不同放射性浓度的89Sr对乳腺癌MCF-7细胞死亡模式的影响及其可能机制。方法用不同放射性浓度的89Sr在体外培养人高转移性乳腺癌MCF-7细胞后,MTT比色法测定细胞增殖抑制率;用流式细胞仪测试、分析培养后细胞死亡模式的变化情况;用免疫组化分析其基因表达情况。结果经不同放射性浓度的89Sr诱导后,MCF-7细胞增殖抑制明显,细胞凋亡和坏死并存,在1480kBqml~6660kBqml范围时p53基因表达增强。结论在一定的放射性浓度范围内,MCF-7细胞增殖抑制率和与89Sr的浓度相关,细胞凋亡和坏死并存,且受p53调控。
