肾组织WT1蛋白表达与大鼠阿霉素肾病模型蛋白尿关系的实验研究

摘要 目的观察WT1(Wilms′tumor 1)在大鼠阿霉素肾病模型中的表达,探讨其在蛋白尿发生发展过程中的作用。方法30只SD大鼠随机分为模型组和对照组,建立阿霉素肾病模型,在第2、4、6、8周,应用光镜、电镜观察肾组织的病理改变,BCA法检测24 h尿蛋白排泄量,半定量Western Blot的方法检测大鼠肾皮质中WT1的表达。结果①随着模型组大鼠肾组织病理改变逐步加重及尿蛋白量逐渐增加,肾皮质WT1的表达量亦逐渐增加,第2、4、6、8周分别为对照组的1.5、2.7、3.1、4.3倍;②WT1的表达水平与尿蛋白量之间呈正相关性(P<0.001)。结论阿霉素肾病模型大鼠肾皮质WT1的表达与大鼠蛋白尿的发生发展密切相关。 Objective To investigate the expression of WT1 (Wilms'tumor 1 ) in rats with adriamycin nephropathy, and to explore the possible effect on the development of proteinuria. Methods 30 SD rats, from 140 to 160g, were randomly divided into the nephropathy model group and normal control group. The nephropathy model was established by a single intravenous injection of adriamycin 6 mg/kg while the normal control group received the same volume of 0.9 % sodium chloride solution. At the end of 2nd, 4th, 6th and 8th week after injection, the rats were sacrificed and their kidneys were acquired. The renal histology was observed with light microscope and electronic microscope. The semiquantitative Western Blot was used to detect the levels of WT1. Results (1) The protein levels of WTI in the nephropathy group were 1.5, 2.7, 3.1 and 4.3 times respectively as high as those in the normal control group. (2) The expression of WT1 was positively correlated with the amount of proteinuria (r = 0.95, P 〈 0. 001). Conclusions The increased expression of WT1 was closely associated with the development of proteinuria in adriamycin nephropathy rats,
