摘要
以腐食酪螨和椭圆食粉螨为材料,采用垂直板型聚丙烯酰胺凝胶电泳法,通过对EST、MDH进行比较研究,探讨了粉螨同工酶技术制样的条件。结果表明:使用pH 7.4的0.01M的磷酸缓冲液制备样品实验效果最佳;取样时通常以样品与匀浆液体积比1:10浓度配比,低于此浓度则容易造成酶带缺失;饥饿对实验效果影响不明显,通常无须饥饿处理。
Abstract.. The Sampling condition of acari for the isozymes was studied in EST.MDH of Tyrophagus putrescentiae and Aleuroglyphus ovatus treated in dtfferent ways By means of polyacrylamide gel electrophoresis. The results showed that samples homogenized in 0.01M phosphate buffer, pH7.4 and with 1 ： 10 in concentration with the homogenate could lead to the optimal zymogram, which was slightly influenced by huger.
