无症状乙型肝炎病毒携带者外周血单个核细胞穿孔素基因表达

Expression of Perforin in Peripheral Blood Mononuclear Cells of Asymptomatic HBV Carriers by Quantitative RT-PCR

摘要 目的:检测无症状乙型肝炎病毒携带者(ASCs)与正常对照组消减文库中的差异基因穿孔素(perforin)在相应人群中的表达。方法:分离43例ASCs和41例正常对照者的外周静脉血单个核细胞(PBMC)并提取RNA,做定量RT-PCR检测。结果:穿孔素在HBV携带者中表达水平是正常对照的3.58倍。结论:穿孔素基因在ASCs高表达,对于阐明ASCs的发病机制有重要意义。 Objective To identify the different expression of perforin between asymptomatic HBV carriers (ASCs) and the normal control. Methods Two to five milliliters of venous blood was collected from the subjects of fourty - three ASCs and forty - one the normal controls, peripheral blood mononuclear cells (PBMC) were isolated freshly, RNA was extracted from the PBMC for real time RT - PCR. Results Perforin expression was upregulated by 3.58 fold in ASCs subjects compared to the normal controls by real time RT - PCR. Conclusion Perforin gene was overexpressed in ASCs compared to the normal controls, suggesting it could play an important role in ASCs.
