人脂肪基质细胞体外分离与培养的实验研究 被引量:3

ISOLATION AND CULTIVATION OF HUMAN ADIPOSE-DERIVED ADULT STROMAL CELLS IN VITRO

摘要 目的探讨人脂肪基质细胞体外分离与培养,为其将来应用于组织工程提供基础。方法通过外科手术获得腹部皮下的脂肪组织,进行体外分离培养脂肪基质细胞,并在相差显微镜下每日观察、记录人脂肪基质细胞的生长状况,并用CCK-8测定其生长生活。结果人脂肪基质细胞呈现梭形外观,增殖旺盛,体外容易扩增。结论我们建立了一种自人体脂肪组织分离、培养人脂肪基质细胞简便稳定的方法,为其应用于组织工程提供了基础。 Objective To establish an approach for isolation and cultivation of human adipose- derived adult stromal (ADAS) cells in vitro. Methods The adipose tissue was obtained from the omentum of abdominal surgery patients under the aseptic condition, then it was isolated and cultured in vitro. The phase- contrast microscope was used to observe human adipose- derived adult stromal cells every day and the viability of human adipose- derived adult stromal cells was assessed by CCK- 8. Results Human adipose- derived adult stromal cells appeared as a monolayer of spindle- shaped, fibroblastic cells with active proliferation,and they can proliferated easily in vitro. Conclusion A simple and stable method for isolation and cultivation human adipose- derived adult stromal cells is successfully established , which provides the foundation for future usage of human ADAS cells in tissue engneering.
