摘要 目的:探讨食管癌组织中血管内皮生长因子(VEGF)的表达水平与肿瘤侵袭和转移的关系。方法:应用实时荧光定量RT-PCR及基因芯片技术对15例食管鳞癌组织和相应正常食管黏膜组织进行VEGFmRNA检测。结果:15例食管鳞癌组织中有8例VEGFmRNA上调表达,其平均表达水平,cDNA微阵列和实时荧光定量RT-PCR检测结果分别为(3.82±0.75)和(4.11±0.89);VEGFmRNA的表达与食管癌患者的年龄、性别、肿瘤的部位、大小、形状、病理分化程度等临床病理特征无关,与临床分期、远处转移有关。结论:VEGF在食管癌浸润和转移过程中发挥重要作用,检测肿瘤组织中VEGFmRNA水平对预测食管癌的侵袭和转移具有重要的临床意义。 Aim: To investigate the relationship between the expression of vascular endothelial growth factor (VEGF) mRNA and invasion and metastasis of esophageal cancer (EC). Methods:The primary carcinoma, the corresponding esophageal epithelium tissue in 15 patients with esophageal squamous cell carcinoma (ESCC) were subjected to detect the level of VEGF mRNA using real time RT-PCR and cDNA microarray, respectively. Results : The expression of VEGF mRNA in 8 (53%) out of 15 patients with ESCC were up-regulated, and the levels were (3.82 ± 0.75) using eDNA chip, and (4.11 ± 0.89) by real time RT-PCR. High level of expression of VEGF mRNA were significantly associated with TNM stages and distant metastases (P 〈 0.01 ) , but not with the sex, age of the patients, and location, size,differentiion degree of EC. Conclusion: VEGF may play an important role in the invasion and metastasis of ESCC. VEGF mRNA assay may be used to predict invasion and metastasis of ESCC.
