食管鳞癌组织中MET原癌基因mRNA的表达 被引量:1

摘要 目的:探讨食管鳞癌组织中原癌基因METmRNA的表达。方法:应用实时荧光定量RT-PCR及基因芯片技术对15例食管鳞癌组织(高分化4例,中分化5例,低分化6例)和相应正常食管黏膜组织进行METmRNA检测。结果:cDNA微阵列和实时荧光定量RT-PCR技术检测结果一致率达100%。15例食管鳞癌组织中有12例METmRNA为上调表达。METmRNA的表达与患者的年龄、性别、肿瘤的部位、大小、肿瘤进展程度等临床病理特征无关(P>0.05),但高、中分化食管癌组织METmRNA表达水平与低分化组比较,差异有统计学意义(P<0.05)。结论:MET原癌基因表达水平与食管鳞癌细胞分化相关。 Aim : To investigate the relationship between the expression of MET proto-oncogene (MET) and cells differentiation in esophageal squamous cell carcinoma (ESCC) tissue. Methods:The expression of MET mRNA in 15 cases of human ESCC including 4 cases of well differentiation,5 cases of moderate differentiation and the normal esophageal epithehum tissue and 6 cases of poor differentiation, was detected by cDNA microarray and real time RT-PCR, respectively. Resuits: MET gene was overexpressed in ESCC compared with normal esophageal epithelium in 12 out of 15 cases (80%). Moreover, the overexpression of MET mRNA was more often observed in well/moderately differentiated ESCC than that in poorly differentiated ESCC (P 〈 0.05). Conclusion : The mRNA overexpression of MET proto-oncogene is correlated with tumor differentiation in ESCC.
