摘要
目的:探索从中华眼镜蛇蛇毒中快速分离提纯磷脂酶A2(PLA2)的方法方法:应用ARG-CY仿生配基,用亲和层析一步法从中华眼镜蛇蛇毒粗毒中纯化PLA2,用SDS-PAGE进行纯度鉴定及相对分子质量测定,并测定其溶血活性。结果:纯化后获得的PLA2电泳图谱呈单一区带,相对分子质量为14000,活性为182μmol/(min·mg)。产物具有溶血活性。结论:仿生亲和层析法提纯LPA2操作简单,纯度高,为一高效实用的方法。
Aim ： To isolate and purify the phospholipase A2 （ PLA2 ） from the Chinese cobra venom. Methods： PLA2 was purified by affinity chromatography. Purified PLA2 was identified and the molecular weight was estimated by SDSPAGE, and its enzymatic activity was tested. Results： Purified PLA2 presented a homogeneous strip on the electrophoresis with strong activity of 182μmol/（ min · mg） and relative molecular weight of 14 000. Conclusion： It is an efficient and simple method for PLA2 purification.
