摘要:通过基因工程方法表达了重组CGAI.76、CGA18-76、CGA18.66和CGA31—76,并利用孔穴琼脂扩散法检测对大肠杆菌和枯草杆菌的抑制作用。结果表明:CGAN端片段对枯草杆菌和大肠杆菌表现出不同的抑制作用;CGA1-76抑菌作用最强,抑菌圈直径达到22mm,而CGAl8.66和CGA31-76的抑菌圈直径不到CGA1-76的一半。CGA18-66没有抑菌圈。因此,推测CGA抗细菌活性中心可能在CGAN端1~17个氨基酸区域。
To study the antibacterial activity of derived fragments of Chromogranin A N-terminal,recombinant CGA1-76,CGA18-76,CGA18-66 and CGA31-76 were expressed by gene engineering.Agar diffusion test was used to test the antibacterial activity of them to Escherichia coli and Bacillus subtilis in this paper.The results show that CGA N derived fragments have different antibacterial activities.CGA1-76 shows highest antibacterial activity,and the diameter of inhibition zone of CGA1-76 is 22mm.While the diameter of inhibition zone of CGA18-66 and CGA31-76 is less than half of it,and there is no inhibition zone of CGA18-66.A conclusion could be drawn that the antibacterial activity domain of CGA perhaps exists in 1~17 amino acids fragment of CGA N terminal.
