摘要
目的研究姜黄素诱导白血病细胞系NB4、K562、THP-1凋亡的分子途径。方法培养上述3种细胞系,用不同浓度的姜黄素(25,12.5,6.25,3.125,0μmol/L)作用24 h和25μmol/L姜黄素作用不同时间(0,12,24,48 h),流式细胞仪测定细胞凋亡率及Fas抗原(CD95)表达,Western blot测定凋亡蛋白Caspase-8、Caspase-9的表达变化。结果①姜黄素以时间和浓度依赖的方式诱导白血病细胞系NB4、K562、THP-1凋亡,25μmol/L姜黄素作用48 h凋亡细胞超过50%。②25μmol/L姜黄素作用24 h后Fas抗原表达明显升高(P<0.01)。③25μmol/L姜黄素作用24 h后凋亡蛋白Caspase-8、Caspase-9的表达明显增高(P<0.01)。结论姜黄素诱导白血病细胞凋亡涉及死亡受体参与的外源性途径和线粒体参与的内源性凋亡途径。
Objective To investigate the molecular pathway of apoptosis of myleocytic leukemia lines NB4, K562 and THP 1 cells induced by curcumin. Methods After the cells were treated with curcumin at different concentrations (25, 12.5, 6.25, 3. 125, 0 μmol/L) for various time (0, 12, 24, 48 h), flow cytometry was used to determine the rate of apoptosis of cells. After the cells were treated with curcumin at 25 μmol/L for 24 h, flow cytometry was used to determine the expression level of Fas, and Western blot was performed to determine the expression of Caspase-8 and Caspase-9. Results ① Curcumin could induce the apoptosis of NB4, K562 and THP-1 cells in a time- and close-dependent manner. After the cells were treated with curcumin at 25 μmol/L for 48 h, the rate of apoptosis of cells was over fifty percent. ② Fas level showed remarkable increase (P〈0.01) after above cells were treated with 25 μmol/L curcumin for 24 h. ③ The apoptosis proteins of Caspase-8 and Caspase-9 were also increased obviously (P〈0.01) after the cells were treated with 25 μmol/L curcumin for 24 h. Conclusion The molecular pathway of apoptosis of myleocytic leukemia cell lines induced by curcumin was concerned with death receptor and mitochondria.
