TRF-1和Tankyrase 1 mRNA在口腔鳞癌细胞中的表达

摘要 目的研究端粒相关调控蛋白端锚聚合酶-1(Tankyrase 1)和端粒重复序列结合因子-1(TRF-1)在口腔颌面部鳞癌细胞中的表达,初步探讨Tankyrase 1对端粒酶活性调控的机制。方法应用组织总RNA抽提技术和实时定量PCR技术进行口腔颌面部肿瘤组织mRNA表达水平的检测,比较了人口腔颌面部鳞癌细胞、良性肿瘤细胞及正常口腔黏膜细胞中Tankyrase 1和TRF-1基因表达的差异及相关性。结果TRF-1 mRNA在口腔颌面部鳞癌细胞、良性肿瘤细胞中的平均表达量显著降低,与正常口腔黏膜细胞对照组相比,差异均具有极显著性意义(均P<0.01),但鳞癌细胞组与良性肿瘤细胞组相比,差异无显著性意义(P>0.05);Tankyrase 1 mRNA的表达在鳞癌细胞组分别高于良性肿瘤细胞组和正常对照组,差异均有显著性意义(均P<0.05),而良性肿瘤细胞组与正常对照组相比,差异无显著性意义(P>0.05)。结论端粒酶活性正向调控因子Tankyrase 1以及逆向调控因子TRF-1在口腔颌面部鳞癌细胞中的表达异常,可能是引起口腔颌面部鳞癌细胞端粒酶活性增高的因素之一。 Objective To study the biological significance of telomeric repeat binding facto-l (TRF-1) and Tankyrase 1 expression in oral squamous carcinoma and the mechanism of Tankyrase 1 regulating telomerase activity, Methods By using tissue total RNA extract and quantitative real-time PCR techniques, the expression of TRF-1 and Tankyrase 1 mRNA was detected in oral squamous tumor tissues, The difference of Tankyrase 1 and TRF-1 mRNA expression in oral squamous cancer cells, oral benign tumor cells and normal oral mucous membrane cells was compared and their correlation was analyzed. Results In the human oral squamous carcinoma cells and oral benign tumor cells, the expression of TRF-1 was significantly decreased as compared with that in the normal oral mucous membrane cells (all P〈0.05), but there was no significant difference between oral squamous cancer cells and oral benign tumor cells (P〉0.05), The expression of Tankyrase 1 mRNA in oral squamous carc- er cells was significantly higher than that in oral benign tumor cells and normal oral mucous membrane cells (both P〈0.05), but there was no significant difference between oral benign tumor cells and normal oral mucous membrane cells (P〉0. 05), Conclusion The abnormal expression of TRF-1 and tankyrase 1 might be one of the factors resulting in the increased telomerase activity in oral squamous cancer cells.
