摘要
目的探讨多西紫杉醇对宫颈癌细胞系Hela的细胞毒性和放射增敏作用。方法MTT法检测多西紫杉醇对Hela的细胞毒性以及1.0nM多西紫杉醇对Hela不同分割放疗方式的增敏效果;克隆形成实验分析不同浓度多西紫杉醇对Hela细胞生长抑制的影响,多靶单击拟合模型方程测定的各组细胞放射增敏比,评价增敏效果。结果0.1 ̄20.0nM多西紫杉醇对Hela细胞的细胞毒性有明显剂量和时间依赖性,20.0nM以上浓度及药物作用超过48h多西紫杉醇的细胞毒性并无明显增加。1.0nM和5.0nM多西紫杉醇放射增敏比分别为1.26和1.69。1nM多西紫杉醇对2.0Gy/次、1.0Gy/次(×2)、0.5Gy/次(×4)分割放疗组的放射增敏比分别为:2.03、2.51、3.57;多西紫杉醇增敏比分别为:1.41、1.82、2.92,每组中放射增敏比较多西紫杉醇增敏比高。结论多西紫杉醇对宫颈癌细胞系Hela有良好的细胞毒性和放射增敏作用,对低剂量分割放射的增敏作用更强。
[Objective] To investigate cytotoxic and radiosensitizing effect of docetaxel on cervical carcinoma cell line Hela. [Method] Cytotoxic effect of docetaxel on Hela cell lines and radiosensitizing effect of 1.0 nM docetaxel combining different low dose fractionated radio therapy was evaluated using MTY assay. Inhibitory effect of different concentration of docetaxel on Hela cell line growth was analyzed using clony formation test, and radiosensitizing effect through sensitizing enhancement ratio (SER) of different group cell line was measured with multi-target singlehit model. [Result] 0.1-20.0 nM docetaxel had obvious dose and time dependent Hela cell line cytotoxic effect, and when concentration over 20.0 nM and treatment time longer than 48 hours, docetaxel shows no more cytotoxic effect. SERs of 1.0 nM and 5.0 nM docetaxel were 1.21 and 1.69, respectively. SERs of 1.0 nM docetaxel combining 2.0 Gy/time (×l), 1 Gy/time(×2), 0.5 Gy/time(×4) fractionated radio therapy groups were 2.03, 2.51, 3.57. Docetaxel enhancement ratios were 1.41, 1.82, 2.92, respectively. [Conclusion] Docetaxel has significant cytotoxic and radiosensitizing effect on cervical carcinoma cell line Hela, especially combining low dose fractionated radio therapy.
