异位子宫内膜间质细胞中芳香化酶的表达及意义 被引量:1

Expression and significance of aromatase in ectopic endometrial stromal cells

摘要 目的探讨芳香化酶在异位子宫内膜间质细胞中的表达情况及其意义。方法体外培养正常和异位子宫内膜间质细胞,采用RT-PCR技术检测其中芳香化酶的表达,发光免疫法检测雌二醇(E2)的表达,流式细胞仪检测凋亡细胞。结果正常子宫内膜间质细胞不表达芳香化酶及E2,细胞凋亡率为7.52%±0.62%;异位子宫内膜间质细胞芳香化酶mRNA为0.89±0.04,E2为(77.24±10.45)pmol/L,细胞凋亡率为1.20%±0.9%,其细胞凋亡率明显低于正常子宫内膜间质细胞(P<0.05)。结论异位子宫内膜间质细胞可以利用芳香化酶合成内源性E2,从而促进细胞增殖。 [Objective] To investigate the expression and significance of aromatase in the ectopic endometrial stromal cells. [Methods]The normal and ectopic endometrial stromal cells were cultured in vitro,the expression of aromatse was measured by RT-PCR,the expression of estradiol was determined by immunofluorometry ,the cellular apoptosis was tested by flow cytometry. [Results] The normal endometrial stromal cells expressed neither aromatase nor estradiol,the cellular apoptosis rate was 7.52±0.62 %. The aromatase mRNA of ectopic endometrial stromal cells was 0.89±0.04,the estradiol was (77.24±10.45) pmol/L ,the celluliar apoptotic rate was 1.20%+0.9%, the apoptotic rate of ectopic endometrial stromal cells was lower than that of normal endometrial stromal cells significently (P〈0.05). [Conclusion] The ectopic endometrial stromal cells can synthesize endogenous estradiol by aromatase,and promate the celluliar proliration.
