深低温保存小鼠卵巢的移植和原始卵泡体外培养

摘要 将新生小鼠的新鲜和深低温保存的卵巢移植到切除卵巢的受体鼠肾被膜下,14d后从移植的卵巢中分离生长中的囊前卵泡,进行体外培养,12d后加入人绒毛膜促性腺激素(hCG)诱导卵子成熟。同日龄小鼠卵巢作为对照。结果显示,移植的新鲜卵巢和深低温保存卵巢中获得囊前卵泡数量较对照组低,卵泡的存活率、发育率和卵子直径等也有差异(P均<0.01);经hCG体外诱导成熟后,新鲜移植、深低温保存后和对照组极体突出卵子的百分率分别为42%、39.8%和61.5%,移植卵巢显著低于对照组(P<0.01)。表明小鼠原始卵泡的成熟可以通过体内移植和体外培养结合的方法实现,深低温保存后卵巢卵泡存活率较高,发育成熟能力良好;但与新鲜卵巢和同龄小鼠卵巢卵泡相比,深低温保存组原始卵泡的数量少、发育能力有一定程度降低。
