摘要
目的探讨白细胞介素4受体(IL-4R)基因α链Arg551Gln基因多态性与支气管哮喘(哮喘)发病及血浆总IgE水平的相关性。方法用等位基因特异性聚合酶链反应(Allele-speccifc PCR)检测哮喘与对照组位点多态性,用酶联免疫分析法(ELSIA)测定哮喘与对照组血浆总IgE水平。结果IL-4Rα链Arg551Gln位点等位基因在哮喘组及对照组间分布的差异有显著性(χ2=6.00,P<0.05),等位基因Arg携带者其哮喘患病危险性较非携带者高1.72倍。三种基因型的分布在两组间的分布有显著性差异(χ2=6.00,P<0.05)。哮喘组Arg/Arg基因型血浆总IgE水平高于Arg/Gln及Gln/Gln基因型,Arg/Gln基因型血浆总IgE水平高于Gln/Gln基因型;而对照组三种基因型血浆总IgE水平两两之间差异无显著性。结论IL-4Rα链Arg551Gln基因多态性是影响哮喘发病的重要因素。
Objective To investigate the correlation among IL-4R α Arg551Glngenc locus polymorphism and asthma and the levels of totle plasm IgE. Methods Allele-specific polymerase chain techniquers were used to determined the Arg551Gln loci polymorphism in asthma and control groups, and the patients's plasm total IgE levels were determined by ELISA. Results The frequency of allele of 551 locus had significant difference between two groups (X^2=6.00, P〈0.05). Allele Arg was associated with asthma, OR=72. The three genetype distribution had significant difference between two groups (X^2=6.00, P〈0.05). In the asthma group, the total plasm lgE levels of subjects of Arg/Arg were higher than those of Arg/Gln and Gln/Gln. The total ptasm IgE levels of Arg/Gln were higher than those of Gln/Gln, However in the control group, the totatptasm IgE levels had no significant differene between subjects of each genetype, Conclusion The polymorphism of IL-4Rα Arg551Gln may be an important factor in asthmatic morbidity.
