摘要
应用改良的2×CTAB法提取到了高质量的11种杜鹃属植物总基因组DNA,可用于杜鹃属植物RAPD分析;用上海生工71个10碱基引物(从S10到S80)筛选到18个具有扩增多态性的引物;应用NEI-LI公式、Prob iosys1.0软件和UPGMA法(配对除权法)进行聚类分析,得到的相似性系数聚类图与传统形态分类一致,为基于DNA分子水平深入探讨杜鹃属植物种间的亲缘关系和系统位置奠定了良好的基础。
The high quality genome DNA of 11 Rhododendron L. was extracted by using the improved 2 × CTAB method, which could be used for RAPD analysis. Moreover 71 kinds of primers (from S10 to S80) from Shanghai Sangon were screened, 18 kinds of amplified polymorphism primers were obtained. Finally, UPGMA method was used to carry out cluster analysis by NEI - LI formula and Probiosysl. 0 software. Its result was basically identical with morphologic taxonomy, which was laid sound foundation for probing into the interspecies relatives and the location of the system of Rhododendron L. based on the DNA molecular level.
