烧伤后血清触珠蛋白对吞噬细胞功能的影响

EFFECT OF SERUM HAPTOGLOBIN ON PHA6OCYTE FUNCTION AFTER BURN INJURY

采用氰化高铁血红蛋白亲和层析法从烧伤血清纯化ＨＰ，并在体外观察纯化ＨＰ对正常小鼠ＰＭＮ释放Ｈ_２Ｏ_２和腹腔Ｍ 抗原提呈功能的影响。结果表明，相当于烧伤血清ＨＰ含量的纯化ＨＰ对正常小鼠ＰＭＮ释放Ｈ_２Ｏ_２和腹腔Ｍ 抗原提呈功能具有显著的抑制作用；而相当于正常血清ＨＰ含量的纯化ＨＰ对ＰＭＮ释放Ｈ_２Ｏ_２和Ｍ 抗原提呈功能均无明显影响。提示烧伤后血清ＨＰ浓度的异常升高可能通过抑制ＰＭＮ的杀菌功能和Ｍ 的抗原提呈功能而参与了烧伤后机体特异性免疫和非特异性免疫功能紊乱的发生机制。

The present study was intended to investigate the effect of haptoglobin,purified from the serum of burned patients by affinity chromatography,on normal murine phagocyte function so as to clarify the role of serum hapoglobin in postburn immunosuppression. The results showed that purified haptoglobin,at the concentration which was equvilent to its serum level detected in burned patients,markedly inhibited the release of hydrogen peroxide by PMNs and the antigen presenting function of normal peritoneal macrophages,whereas it had no significant immunosuppressive activity at the concentration which was equvilent to its level detected in normal serum. According to the results reported here,it is suggested that extraordinary increase of serum haptoglobin concentration following burn injury probably play a role in the pathogenesis of postburn immunosuppression by suppressing specific immune and nonspecific immune.
