摘要
采用高效液相色谱法建立不同季节野葛根的指纹图谱,为野葛根的质量控制提供标准,为这一药食两用资源的合理采收和开发利用提供实验依据.采用的色谱条件为:Agilent1100高效液相色谱仪,Zorbax XDB C18色谱柱,甲醇-水梯度洗脱,流速为0.5 mL/min,检测波长为250nm,柱温25℃.结果显示,野葛根中所含黄酮类成分均得到了很好的分离,指纹图谱表明存在10个共有峰,各共有峰相对峰面积的RSD<5%,共有峰总面积占总峰面积的平均百分比为95.68%,RSD为0.02%;共有峰总面积百分比平均值和葛根素含量平均值均以春季最高.此方法可作为控制野葛根质量的标准,采收野葛根的最适宜季节为春季.
Fingerprint analysis of Pueraria Lobata (Willd) Ohwi with different seasons using high performance liquid chromatography (HPLC) could provide a method of quality control for Pueraria Lobata (Willd) Ohwi and an experimental basis for harvesting the Pueraria Lobata (Willd) Ohwi. The samples were analyzed by HPLC on an Zorbax XDB-C18 column,with mixture of methanol and water as mobile phase in a gradient mode at flow rate 0.5mL/ min and detection at wavelenth of 250 nm, column temperature of 25 ℃. The fingerprint of Pueraria Lobata (Willd) Ohwi was obtained and showed 10 common peak. RSD of relative area of the common peak is less than 10%, Percentage of total area of the common peak is 95.68%, RSD is 1.22% ;The highest percentage of common peak area and content of puerarin were 94.73% and 4.01% in spring. The method could be used in quality control of Pueraria Lobata (Willd) Ohwi. The best season to harvest Pueraria Lobata (Willd) Ohwi is in spring.
