射干有效成分的反相高效液相色谱测定 被引量:18

摘要 用反相高效液相色谱法,36%~60%甲醇为流动相,对66个产地的射干和3种鸢尾中5种异黄酮类化合物,即鸢尾甙、野鸢尾甙、鸢尾甙元、野鸢尾黄素和次野鸢尾黄素进行了分离和含量测定。根据测定结果,对其质量进行了评价。结果表明,川射干含量最高为13.3%,质量较好;混淆品扁竹根含量很低为0.18%,质次,不宜药用。 A reversed phase high performance liquid chromatographic method for thequantitative determination of the five isoflavones(tectoridin,iridin,tectorigenin,irigenin andirisflorentin)in the rhizome of Belamcanda and Iris was developed.The column was packed withTSK gel ODS 80 TS(150 mm×4. 6 mm).Methanol-water(36%~60%)was used as mobile phase.The method was rapid,sensitive, precise and has good reproducibility.The five isoflavones in the crude drngs were completely separated within 40 min,Recoveries ofthe five constituents were 98. 41 %~101. 93 %, The crude drugs were analysed as follows:Thepowdered sample was refluxed with methanol for 6 hours. The extract was transferred into a 2 mlvolumetric flask and diluted to mark with methanol.A definite amount of this sample solution wasinjected into RP-HPLC.The contents of the five constituents in the rhizomes of Belamcanda and Iriswere calculated from the relevant peak heights or peak areas.The analytical results of determination for various species and habitats are shown in Tab 2~5.
