摘要 为探讨大豆甙元磺酸钠对ICR小鼠抑郁性行为的影响,采用不同剂量大豆甙元磺酸盐(50、75、100、1251、50 mg/kg)每天腹腔给药(i.p.),对照组为同体积的生理盐水(i.p.),阳性对照组为同体积的盐酸氟西汀(10 mg/kg).给药期间或结束后用强迫游泳抑郁行为测试方法(Forcedswimming test,FST),观察并记录各组游泳不动时间(s),并进行统计分析.结果显示,雄性ICR小鼠第7 d测试结果与对照组相比,显著降低游泳不动时间(P<0.05);第14天测试75 mg/kg组和125 mg/kg组与对照组相比能够显著缩短不动时间(P<0.05);连续给药18 d后75 mg/kg组和100 mg/kg组与对照组相比分为显著(P<0.05)和极显著缩短不动时间(P<0.01);阳性对照(10 mg/kg)氟西汀组只有在给药18 d后显著降低雄鼠的不动时间(P<0.05).同时,各种给药剂量对于正常雌鼠或摘除卵巢雌鼠的游泳不动时间均没有显著影响.结果表明,大豆甙元磺酸盐可以降低小鼠的游泳不动时间从而发挥抗抑郁作用,而且该作用与药品的剂量、给药持续时间以及小鼠的性别密切相关,在雌、雄鼠之间的作用差异可能与大豆甙元磺酸盐的雌激素样作用有关. To observe the effects of daidzein sulfonic sodium on the depressed benaviors of male & female ICR mice, the following method has been carried out. 3'-sodium sulfonate daidzein (SSD) was injected for mice in different dose (50, 75, 100, 125, 150 mg/kg), and mice in the control group were injected normal saline in the same volume as SSD, and mice in the positive-control group were injected fluoxetine (10 mg/kg) in peritoneal cavity. According to the depressed behavior way of forced-swinming test, the immobility times of mice in every group were recorded. Based on the statistics and analysis of the data, the results show when the male mice were used as test animal, in comparison with the control group at the 7th day's test, SSD reduced immobility time (IT) significantly for dose 75 mg/kg (P〈0.05) ; and at the 14th day's test, SSD reduced IT significantly for both dose of 75 mg/kg and 125 mg/kg (P〈0.05) ; After 18 days' injecting drug, in comparion with the control group, SSD reduced IT significantly for dose of 75 mg/kg (P〈0.05) and 100 mg/ kg (P 〈0.01), and the positive-control group that injected fluoxetine reduced IT significantly (P 〈0.05) for dose of 10 mg/kg. However, there is not any effect on female mice's IT in every experiment condition; It suggested that antidepressant effect of SSD depends on the dose of drug, the time of injecting drug, and the sex of mouse. The different effect on the male and the female mice may be related to estrogen-like activity. However, other biological ways don't be exclude yet. Therefore the further research of the mechanism is necessary.
