摘要
目的:了解天津地区宿主鼠间感染的汉坦病毒的基因分型。方法:提取经免疫荧光检测证实为汉坦病毒抗原阳性的鼠肺组织的总RNA,经RT-PCR扩增汉坦病毒S基因片段,其产物纯化后与T载体连接并进行序列分析。结果:6份鼠肺标本RT-PCR产物均为阳性,序列分析表明J16为HTN型,B11、BC3、D17、H4、J37均为SEO型,且属于同一亚型。结论:天津地区鼠中存在汉坦病毒主要为HTN型和SEO型。
Objective: To investigate the hantavirus genotype infected among host rodents in Tianjin area. Methods: The total RNA extracted directly from rodent lung tissues which was determined to be hantavirus antigen positive by IFA methos and the hantavirus S gene segment was amplified by RT-PCR methods and the product was purified and cloned into T vector, then the sequence was analyzed. Results: The RT-PCR product rodent results of six specimens were all positive. The sequence analysis indicates that J16 was HTN type. Bll,BC3,D17,H4 and J37 were SEO type and they were all the same subtypes. Conclusion: Hantavirus infecting among rodents mainly were HTN type and SEO type in Tianjin.
