摘要
目的比较核素药物锝-99-亚甲基二膦酸钠(^(99)Tc-MDP,俗称云克)和中药速效骨伤愈合剂(俗称“骨王”)治疗骨质疏松动物模型的疗效,研究其在骨承受力等方面的变化。方法采用盐酸地塞米松肌内注射方法诱导兔骨质疏松模型,实验设有正常对照组、骨质疏松模型对照组以及骨质疏松模型分别按成人常规剂量比例给予^(99)Tc-MDP和速效骨伤愈合剂的治疗组,治疗时间为15周。治疗前、后进行实验室X线、CT摄片及核素骨骼显像,取第3、4腰椎和股骨分别进行病理细胞学、骨形态计量分析,并行骨密度和耐压骨力学试验。结果^(99)Tc-MDP治疗组与骨质疏松模型对照组比较,除X线、CT摄片显示的变化不明显外,在病理细胞学、骨形态计量、骨密度、耐压骨力学方面的差异均有显著性(P<0.01),核素骨骼显像显示骨组织摄取放射性提高;速效骨伤愈合剂治疗组在骨形态计量方面提高较明显,耐压骨力学测试结果较^(99)Tc-MDP治疗组差,^(99)Tc-MDP治疗组与正常对照组的差异略有显著性。结论核素药物^(99)Tc-MDP对兔骨质疏松模型的治疗有较好的疗效,虽然中药速效骨伤愈合剂治疗骨质疏松动物模型有一定疗效,但关键的耐压骨力学的变化较小,可能速效骨伤愈合剂需要在较长的时间内逐渐对骨组织的增殖功能起促进作用。
Objective Compare the curative effect of ^99Tc-MDP and traditional Chinese medicine "Gu Wang" in treating osteoporosis, and study the bone handling capacity of the animal model. Method The osteoporotic rabbit model was constructed by intramuscular injection of dexamethasone hydrochloride. The animals were divided into normal control group and osteoporotic model group with application of ^99Tc-MDP and "Gu Wang" respectively, in a course of 15 weeks. Before and after therapy, X-ray, CT, skeletal imaging were performed. The 3rd and 4th lumbar vertebrae and femur were studied with cellular pathology, bone morphometry analysis, bone density and pressure holding test. Results In the ^99Tc-MDP group, there was obvious improvement in the above parameters, including the skeletal nuclide imaging showed improved radioactivity bone absorption. In the "Gu Wang" group, improvement was seen in bone morphometry, but the pressure holding capacity was worsened than that of the for mer. But ^99Tc MDP group showed a slight difference with control group. Conclusion ^99Tc-MDP shows perfect curative effect in treating rabbits osteoporotic model. The traditional Chinese medicine "Gu Wang" shows certain effectiveness in the same treatment, but without obvious change on the pressure holding capacity of bone, probably requiring longer duration of proliferation of bone tissue.
