白念珠菌蛋白酶对中性粒细胞和淋巴细胞的趋化性研究 被引量:5

Study on Chemotactic activity of Candida Albicans Proteinase for Neutrophils and Lymphocytes

摘要 采用改良 Boyden 室法测定了白念珠菌蛋白酶(Candida albicans proteinase,CAP)对人末梢血白细胞的趋化活性。发现 CAP 对中性粒细胞的趋化活性随浓度增高而增强,在500nmol/L 时达到最大值(P<0.001)。100℃处理 CAP 10分钟后趋化活性显著降低。棋盘法分析发现 CAP 对中性粒细胞不仅有趋化活性,而且有化学激活性。CAP 仅引起淋巴细胞轻微趋化。在3只豚鼠真皮内注射 CAP,3小时后的活检标本中发现真皮血管周围的中性粒细胞平均浸润数为29个/视野,显著多于注射磷酸盐缓冲液的平均3个/视野(P<0.05)。此结果提示,作为蛋白水解酶的 CAP 不仅与菌体侵入机体和增殖有关,同时可诱导末梢血中性粒细胞向感染局部移动。 The chemotactic activity of Candida albicans proteinase toward human peripheral leuko- cytes was examined by a modified Boyden chamber method.It was found that C.albicans proteinase in- duce dose-dependent chemotactic activity for neutrophils with maximal activity at 500 nmol/L(P< 0.001).Chemotactic activity was significantly reduced after 10 min treatment at 100℃.Checkerboard analysis indicated that C.albicans proteinase is chemotactic and activates neutrphils chemically.Mild chemotactic activity was observed for peripheral lymphocytes.C.albicans proteinase injected intrader- really into the skin of 3 guinea pigs,it induced significantly more infiltration of neutrophils(29 cells/ field)around the dermal vessels than did phosphate buffered saline(3 cells/field,P<0.05)at the 3- hour biopsy samples.These results suggest that C.albicans proteinase,which theoretically acts as a hy- drolase for fungal invasion and growth,induces the migration of peripheral neutrophils to the C.albi- cans-infected site.
