摘要
目的:探索电热针对Ⅱ型胶原诱导性关节炎(Collagen-induced Arthritis,CIA)大鼠踝关节滑膜细胞环氧化酶-2 mRNA(COX-2 mRNA)表达的影响。方法:随机将50只Wistar大鼠分为正常组、模型组、电热针高电量治疗组(高电组)、电热针低电量治疗组(低电组)、电热针无电量治疗组(无电组)。Ⅱ型胶原足底皮下注射造模;其中正常组足底假免疫时给予生理盐水。电热针组于致炎后第21 d开始治疗,取双侧“阳陵泉”穴,直刺8 mm,每次20 min,隔天1次,共治疗20次。其中高电组电流强度为40 mA,低电组电流强度为20 mA。治疗结束后,各组取左踝关节组织,以原位杂交法检测踝关节滑膜细胞COX-2 mRNA表达。结果:造模后,各造模动物的踝关节肿胀程度比正常组显著增加(P<0.01);电热针治疗后,各治疗组与模型组比较踝关节肿胀程度有显著减轻(P<0.01)。模型组踝关节滑膜COX-2 mRNA表达较正常组显著增高;与模型组相比,高电组踝关节滑膜COX-2 mRNA表达显著降低(P<0.05),低电组、无电组与模型组则无明显差异。结论:电热针具有抗炎作用,仅有高电量电热针治疗可以降低CIA大鼠踝关节滑膜细胞COX-2 mR-NA的表达。
Objective: To observe the effect of electrothermo-needle(ETN) treatment on Cyclooxygenase (COX)-2 mR NA expression osteosynovial cells in collagen-induced arthritis (CIA) rats. Methods: A total of 50 Wistar male rats ( 180 - 200 g) were randomized into normal control, model, higher electric current (40 mA) ETN, low-current (20 mA) ETN, no-current (0 mA) ETN groups, with 10 cases in each group. Arthritis model was established by injecting type- Ⅱ collagen ( 100 μg) into the rats' subcutaneous tissues of the bilateral hind-paws. ETN was applied to bilateral “Yanglingquan” (GB 34) once every other day with 20 min each time and 20 times altogether. The rats' ankle-joint swelling degree (intensity of pressure) was detected by using joint volume detector. COX-2 mRNA expression of osteosynovial cells of the left ankle joint was assayed by using in situ hybridization technique. Results: In model group, the intensity of pressure of the rats' bilateral ankle joints increased significantly in cornparison with that of rats in normal control group, while the intensity values of the ankle joints in high-current ETN, low-current ETN and no-current ETN groups were remarkably lower than that of model group (P〈 0.01 ); no significant differences were found among high-current ETN, low-current ETN and no-current ETN grips in the swelling degree (P〉0.05), suggesting that after ETN, the joint swell decreased significantly. Compared with normal group, there was a significant increase in COX-2 mRNA expression in model group (P〈0.05), while in comparison with model group, COX-2 mRNA expression was downregulated significantly in joint synovitis in high-current ETN group ( P〈 0.05). There were no significant differences between low-electric current and model groups, and between model and no-electric current groups in COX-2 mRNA expression (P 〉0.05). Conclusion: Electrothermoneedle has a favorable effect in anti-arthritis,and high intensity ETN can downregulate the expression of COX-2 mRNA in joint synovitis in arthritis rats.
