摘要
目的:研究转化生长因子(TGF-β2)和结缔组织生长因子(CTGF)在滤过泡瘢痕化中的作用。方法:用不同浓度的TGF-β2刺激HTF,然后用RT-PCR和免疫细胞化学法分别检测CTGF和Fn的表达,用MTT法检测细胞的增殖。结果:TGF-β22μg/L到5μg/L范围有剂量依赖性地促进HTF的细胞增殖,CTGF的表达和Fn的合成的趋势(P<0.01),但0.5μg/L,1μg/L与正常对照组对细胞的增殖作用差别无显著性,5μg/L与10μg/L增强细胞的增殖作用也无显著性差异(P>0.05)。结论:TGF-β2能够明显地促进HTF细胞增殖,并增强其下游介质(CTGF)的表达和合成细胞外基质-纤维连接蛋白(fibronetin,Fn)。
AIM: To investigate the influence of transforming growth factor-13 2 and connective tissue growth factor on the human Tenon's capsule fibroblasts.
METHODS: The proliferation of HTF treated by different concentration of TGF-β 2 was detected by using MTT colorimetry, and the expression of CTGF and fibronectin was detected by RT-PCR and immunocytochemistry assay respectively. Imaging and statistical analysis was used to evaluate the effect of transforming growth factor.
RESULTS: MTT method: the induction of TGF-β 2 (2, 4 and 5μg/L) showed a promotion on the proliferation of HTF and the expression of CTGF and fibronectin (P〈0.01),while 0.5μg/L and 1μg/L group vs. normal group, 5μg/L group vs 10μg/L group had no significant difference on the proliferation of HTF (P〉0.05).
CONCLUSION: TGF- β2 can enhance the proliferation of the HTF and induce the expression of CTGF and Fn in a dose-dependent manner, inferring that CTGF may be an important cytokine in scarring after glaucoma filtering surgery as a downstream mediator of TGF-β 2 action.
