摘要
[目的]研究磨损颗粒刺激单核细胞thp-1后,细胞分泌TNF含量和刺激后条件培养基引起成骨细胞的凋亡。[方法]L929细胞测活和ELISA方法检测单核细胞TNF-α的分泌和分泌的量;TUNEL方法检测磨损颗粒条件培养基对成骨细胞的凋亡作用。[结果]不同种类磨损颗粒加入thp-1后TNF-α的结果用方差分析比较, 3组数据之间有显著性差异(P<0.05)。将不同颗粒的100 ng/ml组和10 ng/ml组分别两两比较,两组之间结果有显著性差异(P<0.05),100 ng/ml比10 ng/ml TNF值高。[结论]磨损颗粒PE、Co-Cr-Mo、Ti-6Al-4V刺激体外培养单核细胞thp-1分泌TNF-α,3种磨损颗粒之间有显著差异(P<0.05)。磨损颗粒PE、Co-Cr-Mo、Ti-6Al-4V刺激单核细胞thp-1的条件培养基可以引起体外培养成骨细胞凋亡。
[Objective] To evaluate the secretion of TNF-α by monocyte thp-1 stimulated by wear particles and the apoptosis of osteoblasts after cultured in the conditioned culture of wear particles and thp-1. [Methods] L929 and ELISA were used to measure the secretion of TNF-α, TUNEL were used to determine the apopotosis of osteoblasts cultured in the conditioned culture. Student t test were used to analyse the TNF results. [Results] The difference was singnificant among 3 different particle groups, and between 100 ng/ml and 10 ng/ml group. [Conclusion] PE, Co-Cr-Mo and Ti- 6Al-4V particles can stimulate thp-1 to secrete TNF-α, and there are singnificant differences among these 3 particle groups.
