结肠癌原发灶及其淋巴结转移灶中PTEN表达的微阵列研究 被引量:1

摘要 [目的]检测结肠癌原发灶及其淋巴结转移灶中PTEN的表达情况。[方法]采用组织芯片技术,应用免疫组化方法对79例结肠癌原发灶及其淋巴结转移灶标本进行PTEN的表达情况检测。[结果]原发灶与其淋巴结转移灶相比:PTEN在粘液腺癌中的表达强度明显降低(P<0.05),而管状腺癌中其表达无明显差别; PTEN的表达与肿瘤分化程度无关。原发灶及其转移灶中PTEN的高表达率在管状腺癌中均显著高于粘液腺癌。[结论]淋巴结中转移的粘液腺癌细胞表达较低水平的PTEN,其低表达可能有助于结肠癌的淋巴道转移。 [Objective] To examine the expression of PTEN protein in primary foci of colon carcinoma and their lymphnode metastatic loci. [Methods] Immunohistochemical staining of PTEN were performed on two tissue microarrays made from 158 paraffin specimens, including primary colon carcinoma and their lymph-node metastatic loci. [Results] Expreasion level of PTEN protein in metastatic mucoid adenoma was significantly lower than that in primary colon carcinomas (P 〈 0.05). Expression level of PTEN protein has no relationship with the degree of differentiation. High expression rate of PTEN in tubiform adenocarcinoma was significantly higher than that in mucoid carcinoma both in primary focus and lymph-node metastatic loci. [ Conclusion] Mucoid cancer cells in lymph-node metastatic loci express low level of PTEN protein. Low expression level of PTEN protein may conduce to lymph-node metastases of colon carcinoma
