摘要
目的探讨K562/A02细胞株NF-κB活性与P-gp表达的关系。方法在K562/A02白血病耐药细胞株培养中加入NF-κB抑制剂PDTC,观察在不同浓度、不同时间下,细胞株NF-κB活性的变化与P-gp表达的关系。结果在不同浓度、不同时间的PDTC作用下,K562/A02细胞株NF-κB活性与P-gp表达呈正相关的关系,随干预浓度、时间的不断变化,二者均逐渐下降。结论NF-κB调控的白血病细胞抗凋亡机制中有MDR1的参与。
Objective To explore the relationship between nuclear factor-rB (NF-kB) and P -glycoprotein (P-gp) on K562/A02 cells. Methods NF-kB and P-gp of K562/A02 cells cultured with PDTC in different concentrations and time were detected by immunohisochemisty, and the relationship between them was analyzed. Results In K562/A02 cells, the expression of NF- kB and P-gp were positively correlated, and were concentrationdependent and the time dependent. Conclusion The mechanism of NF- kB regulating anti-apoptosis of K562/A02 cells is related with MDR1.
