胆固醇24S-羟化酶与阿尔茨海默病 被引量:3

摘要 阿尔茨海默病(AD)是一种进行性神经变性疾病,65岁以上的痴呆人群中,有一半以上可归因于AD.AD的特征性病理变化有两类:一是以β-淀粉样蛋白(Aβ)为中心的沉积于细胞间的老年斑,二是细胞内的神经原纤维缠结(NFT).迄今为止,AD的病因尚未明了.大量的研究表明,胆固醇的代谢在AD的发病机制中可能起着重要作用,Aβ的产生、聚积及tau蛋白的磷酸化均与胆固醇代谢有关.然而,胆固醇不能透过血脑屏障,在颅内,它主要通过胆固醇24S-羟化酶(CYP46)转化为24S-羟胆固醇,24S-羟胆固醇能够通过转运机制透过血脑屏障,从而保持胆固醇在脑内的动态平衡.
