摘要
目的探讨利用软骨细胞和新型生物材料壳聚糖动物体内构建软骨组织的可行性。方法体外分离培养猪耳软骨细胞,扩增后以5.0×107/ml的终浓度接种于壳聚糖支架并植入猪皮下。标本于8周后取材,行大体观察、组织学及免疫组织化学等相关检测。结果细胞-生物材料复合物在植入猪皮下8周后形成了单一成熟的软骨组织,并保持了支架材料的大小和形状,免疫组织化学染色结果表明其大量表达软骨特异性细胞外基质Ⅱ型胶原。结论壳聚糖是一种良好的生物材料,能够提供软骨细胞生长的三维环境并用于体内构建软骨组织。
Objective To explore the possibility to construct cartilaginous tissue in vivo by using cartilage cell and a new type biomaterial chitosan. Methods Cartilage cells were separated from pig ears and cultured in vitro. After amplification, cells were seeded at a final concentration 5.0 × 10^7 per ml on chitosan frame, which was then implanted subcutaneouly in pig. 8 weeks later, samples were collected and detected with histology and immunohistochemistry. Results Cartilage cellChitosan complex cultured for 8 weeks subcutaneouly formed sigle ripe cartilaginous tissue, and remained the size and shape, the result of immunohisto-chemistiy indicated that a great quantity of Ⅱ -type collagen were expressed, which were specific for the extracellular of cartilage. Conclusion Chitosan was a favourable kind of biomaterial, which could provide 3D environment for the growth of cartilage cells to construct cartilaginous tissue in vivo.
