阳离子脂质体介导BEL-7402、QBC、BXPC-3瞬时转染率比较

摘要 目的:探讨阳离子脂质体瞬时转染消化道肿瘤细胞的差异。方法:采用pEGFP-N1质粒为报告基因,流式细胞仪技术(FACS)来分析阳离子脂质体介导3种细胞的瞬时转染率,设立裸质粒的转染为空白对照组,将转染的细胞在倒置荧光显微镜下观察并摄像;采用MTT法检测3种细胞的倍增时间,并绘制细胞生长曲线;分析细胞倍增时间和阳离子脂质体介导的转染之间的联系。结果:BEL-7402,QBC,BXPC3 3系细胞阳离子脂质体介导的瞬时转染率分别为26.99%、2.25%和30.36%,BEL-7402和BXPC-3同QBC细胞有显著差异(P<0.05);3个细胞系裸质粒的转染率分别为0.22%、0.29%和0.18%,3者之间差异无显著性(P>0.05);BEL-7402、QBC、BXPC-3的倍增时间分别是34.48 h、64.94 h和26.29 h,QBC细胞同BEL-7402及BXPC-3相比差异有显著性(P<0.05)。结论:阳离子脂质体对高度恶性的消化道肿瘤细胞有更好的转染效率。 Objective: To investigate the difference of cation lipofectin-mediated transfection (CLT) among different malignant tumors. Methods: Three cell lines of BEL-7402, QBC, and BXPC-3 were transfected by plasmid pEGFP-N1. The transfection in experimental group was cation lipofectin-mediated, while that in control group without the mediation. The transfection results were detected by using FACS and fluorescent microscopic photo analysis; pEGFP-N1 transfected cells were observed and their growth curves and multiplication times were monitored with MTT method; The relation of multiplication time and transfection efficiency was analized. Results: The CLT efficiency of the three cell lines of BEL-7402, QBC, and BXPC-3 were 26. 99%, 2. 25% and 30. 36% repectively. The CLT efficiency of QBC was significantly lower than those of BEL-7402 and BXPC-3 ( P 〈 0. 05 ). The transfection efficiency of the pEGFP-N1 of the three cell lines in the control group were 0. 22%, 0. 18% , and 0. 29% respectively. No significant difference was found among them ( P 〉 0. 05 ). The multiplication time of Bel-7402, QBC and Bxpc-3 were 34. 48h, 64. 94h and 26. 29h, and the multiplication time of QBC was significantly longer than those of BEL-7402 and BXPC-3 ( P 〈 0. 05). Conclusions: Significantly higher instant transfection efficiency in the malignant carcinoma cells can be achieved with the help of cation lipofectin-mediation.
