摘要 目的:观察血竭提取物对体外培养成纤维细胞增殖的影响,探讨血竭促进创面愈合的机制.方法:将血竭经过氯仿、乙酸乙酯、乙醇回流提取得到的3种提取液,分别加入培养液并进行成纤维细胞的培养,MTT法检测不同血竭提取物在不同浓度以及不同时间点对体外培养成纤维细胞增殖的影响,并绘制最适浓度条件下细胞生长曲线.利用流式细胞仪分析最适浓度培养条件下成纤维细胞的细胞周期变化.结果:血竭乙酸乙酯提取物在0.5—2.0g/L浓度范围内,促进成纤维细胞增殖,且呈剂量依赖性,在浓度为2.0g/L时促进作用最为显著,此条件下处于S期的细胞较对照组增加18.3%(P〈0.01).结论:血竭乙酸乙酯提取物能显著促进成纤维细胞增殖,可能与血竭促进创面愈合的机制有关. AIM: To investigate the effect of Dragon's blood on the proliferation of fibroblast cells in vitro and to explore the mechanism by which Dragon's blood promotes wound healing. METHODS: Human fibroblast cells were cultured in vitro in media containing gradient concentrations of 3 kinds of Dragon's blood extracts (extracted by chloroform, acetidin and alcohol, respectively), which was followed by cell proliferation determination assessed with MTr assay at different time intervals. The growth curve at the most suitable concentration was drawn. The flow cytometry (FCM) was used to determine the changes of cell cycle at this concentration. RESULTS : The 0.5 - 2.0 g/L dilution of Dragon's blood extract by acetidin enhanced the proliferation of fibroblast in a dose-dependent manner. The 2.0 g/L was the optimal concentration of Dragon's blood extract, and it could increase the cell ratio in S-phase of cell cycle by 18.3% of that in control group ( P 〈 0.01 ). CONCLUSION : Dragon's blood extract by acetidin could promote the proliferation of cultured human fibroblast cells in vitro, and it might be beneficial to granulation tissue formation during wound healing.
