穴位埋线对实验性癫痫大鼠海马及大脑皮质γ-氨基丁酸和谷氨酸的影响 被引量:3

Influence of catgut-embedding therapy on GABA and Glu in hippocampus and cerebral cortex in rats with experimental epilepsy

摘要 目的探讨穴位埋线对实验性癫痫大鼠大脑海马及皮质中γ-氨基丁酸(GABA)、谷氨酸(G lu)含量的影响。方法50只大鼠随机分为正常组、模型组、埋线组、针刺组、丙戊酸钠组,经预处理后用青霉素钠腹腔注射造模(正常组除外),造模后90 m in时处死大鼠,迅速取一侧大脑海马和颞叶皮质,以高效液相色谱仪测定海马和颞叶皮质中GABA、G lu的含量,计算GABA/G lu值。结果模型组海马区GABA、G lu的含量和GABA/G lu值均高于正常组(P<0.05,P<0.01),埋线组GABA含量和GABA/G lu值均明显高于模型组(P<0.01)、而其G lu的含量明显低于模型组(P<0.01),且埋线组的影响大于丙戊酸钠组、针刺组,穴位埋线对海马区GABA、G lu的影响比颞叶皮质区明显。结论穴位埋线可通过调节脑内兴奋性与抑制性氨基酸的水平而发挥抗癫痫作用。 Objective To investigate the catgut-embedding therapy on the contents of γ-aminobutyric acid (GABA) and glutaminc acid (Glu) in hippocampus and cerebral cortex in rats with experimental epilepsy. Method To divide randomly 50 rats into the normal group ( group 1 ), model group ( group 2), catgut-embedding therapy group ( group 3 ), acupuncture group ( group 4 ) and valproate sodium group (group 5 ), after 3 days pretreatment, and then the introperitoneal injection of benzylpenicillin sodium was given to different groups ( besides group 1 ) to establish the model. After 90 minutes the rats were killed and the hippocampus and cerebral temporal cortex were taken from one hemisphere of brain. The contents and ratio of GABA and Glu were determined and calculated by HPLC. Result In group 2 the contents and ratio of GABA and Glu were all higher than those in group 1 (P 〈 0. 05, P 〈 0, 01 ), the content of GABA and ratio of GABA and Glu in group 3 were all higher than those in group 2 ( P 〈 0. 01 ), but the content of Glu in group 3 was obviously lower than that in group 2 ( P 〈 0.01 ). The influence of the catgut-embedding therapy in group 3 was stronger than that in group 5 and group 4. The therapy had more significant effect on hippocampus catgut-embedding therapy has the antagonism on inhibiting amino acids level. than on cerebral temporal cortex.
