摘要 目的探讨奥曲肽对角膜新生血管形成及角膜内血管内皮生长因子(vascularendothelial growth factor,VEGF)和色素上皮衍生因子(pigment epithelium derived factor,PEDF)表达的影响。方法制备大鼠角膜碱烧伤新生血管模型,分每日腹腔注射奥曲肽20μg组、30μg组和对照组3组。运用裂隙灯、免疫组化、电镜分别观察新生血管长度及生长速度、2种因子的表达。结果治疗组新生血管生长速度变慢,微血管数量减少,VEGF蛋白表达下降,PEDF蛋白表达上升,具有统计学差异(P<0.05)。结论VEGF和PEDF的表达与角膜新生血管的形成过程密切相关,一定剂量的奥曲肽可抑制角膜新生血管的发生。 Objective To discuss the inhibitive effects of Octreotide on angiogenesis and expressions of vascular endothelial growth factor(VEGF) and pigment epithelium derived factor (PEDF). Methods Alkali burns of corneal model was established as usual, the Wistar rats were equally subdivided into three groups, one group was set as control, the other two received lntraperitoneal injection of Octreotide 20 μg and 30 μg for 2 weeks. The occurrence and development of corneal neovascularization (CNV) were observed by three methods: thslit-lamp microscopy, counting of microvessl by microscopy and immunohistochemical analysis of VEGF and PEDF respectively. Results In the experiment groups, CNV grew more slowly, microvessl number was fewer, VEGF protein was less and PEDF was more than those in the control one. VEGF protein and PEDF protein were temporally and spatially correlated with CNV.Conclusion VEGF and PEDF are intimately correlated during the development of corneal neovascularization, and some dosage of Octreotide can decrease the expression of VEGF and increase the expression of PEDF to inhibit the occurrence of CNV.
